IRCCS San Raffaele Scientific Institute in Milan–Italy, is looking for a young motivated researcher to join the Cell Adhesion Laboratory directed by Prof. Ivan de Curtis. The candidate will work on a project aimed at analyzing molecular–cellular mechanisms implicated in tumor cell invasion, to identify new targets to interfere with the formation of metastases. The project financed by AIRC focuses on the functional characterization of a molecular network that regulates the protrusive activity of invasive tumor cells. The protein network includes the scaffold proteins Liprin‑a1 and ERC1 and regulatory enzymes undergoing liquid‑liquid phase separation to regulate invasive properties of breast cancer cells, promoting turnover of adhesions and invasion. The candidate will investigate mechanisms linking Liprin‑a1 and ERC1 proteins to the organization and function of the migratory activity of cancer cells. Tasks include cloning to prepare wt / mutant constructs, high resolution confocal microscopy on fixed and live cells, biochemical / molecular cell biology approaches including transfections with siRNAs and plasmids for KO (CRISPR / Cas9) of endogenous proteins, and functional assays to assess effects on tumor cell invasion.

Main Activities

Investigate the molecular mechanisms linking scaffold proteins Liprin‑a1 and ERC1 to the organization and function of cancer cell migration and invasion. Apply cloning, live‑cell imaging, transfections, CRISPR / Cas9 knock‑out strategies, and functional invasion assays.

Qualifications

Post‑master degree or post‑doctoral candidate with strong motivation to learn molecular‑cell biology approaches relevant to cancer cell biology. Experience with cloning, transfection, siRNA, CRISPR / Cas9, and live‑cell imaging is desirable. Strong motivation and interest in tumor cell invasion and metastasis research.
